EFFICIENCY OF IMAGE ANALYSIS AS A DIRECT METHOD IN SPORE DIMENSIONS MEASUREMENT

Abstract
This method summarizes calculation of dimensions of different types of micro cells as bacterial, fungal spores, and blood cells. Uni and multicellular organisms like plant tissue, animal, hyphae are included. Any microscopic sample can be examined using the image analysis software Digimizer with photo taken under light microscope. The software can determine dimensions of objects with no need of calibration for an ocular micrometer or stage micrometer. Analyzing image by software was considered a direct method to measure microscopic sample dimensions. It is a calibration based on identified dimension within the prepared sample. In this research, Hemocytometer squares were used as a predetermined dimension to calibrate the software then analyze images. The result exhibited efficiency of the image analysis method in microbiological sample, spore suspension of mushroom Agaricus bisporus, calculating. The method was proven through statistical analysis of the two mentioned methods.
