Abstract

In the present investigation, a protocol was developed for in vitro propagation of Sterculia foetida Linn. using cotyledonary node segment which was selected after screening of different seedling explants. Maximum number of multiple shoots produced from the cotyledonary node when placed on MS medium incorporated with BAP (4 mg/l). Optimum sucrose concentration for multiple shoot formation was 3%. Role of different growth additives like CH, AA, Ad, ME, BE, YE and CM was assessed in enhancing shoot number. The regenerated micro shoots were isolated and cultured on the rooting medium containing various concentrations of IAA, IBA and NAA (0.1-2.0 mg/l). IAA at 2 mg/l was found to be best for highest frequency of root formation from the cut ends of the micro shoots without callus formation. The regenerated plantlets were acclimatized in the field conditions with 40% survival rate.
