Abstract

The present study is focused on induction of multiple shoots from shoot tip explants of cucumber. Shoot tip explants were excised from 7-day-old seedlings and cultured on MS medium fortified with different concentrations of (0.5-2.5 mg/l) BAP alone and/or in combination with 0.5 mg/l KIN for shoot induction. The highest number of multiple shoot regeneration was noticed on MS medium supplemented with 1.0 mg/l BAP produced maximum frequency (89.8 %) with 9.8 shoots/culture. The elongated shoots were excised and cultured on half- strength MS medium containing different concentrations of (0.5-2.5 mg/l) IBA (Indole 3- butyric acid) and (0.5-2.5 mg/l) NAA a-Naphthalene acetic acid) alone and in combination with 0.5 mg/l KIN for rooting. Among the combinations used, IBA (1.5 mg/l) and KIN (0.5 mg/l) combination induced maximum percent of rooting (96.82%) with 8.24 roots/shoot. Well developed plantlets were successfully transferred into plastic cups containing a mixture of soil and sand in the ratio 1:1 and subsequently established in the green house and acclimatized in the field.
