Abstract

Tomato is one of the most important and essential vegetable crops all over the world. The present study was focused on genetic transformation and plant regeneration of tomato (Lycopersicon esculentum cv PKM-1). Transformation was carried out by Agrobacterium tumifaciens harboring binary vector pBI121 containing modified Cry1Ac gene. Three explants namely cotyledonary node, cotyledonary leaves and hypocotyl was used in transformation. After co-cultivation, transformed explants were selected on Kanamycin medium (100 mg/l) and selected explants were cultured for regeneration into shoots. Regenerated shoots were rooted and plants were established. Some of the established plants were tested PCR positive with gene specific primers for the presence of Cry1Ac gene.
