In vitro Plant Regeneration via Somatic Embryogenesis for Cinnamum smophloeum Kaneh

Abstract
A protocol for somatic embryogenesis and plant regeneration from immature leaflets of Cinnamum osmophloeum has been developed. Induction of callus was on woody plant medium (WPM) supplemented with 2,4-D (2 mg/l). A high percentage of embryogenesis (64.7%) and average number embryos (30.5) appeared after primary callus was transferred to WPM medium supplemented with 2,4-D (0.1 mg/l) with 2ip (0.05 mg/l) . The enhanced rate of embryogenesis (85.9%) and average number of embryos (69.6) were achieved (data not shown) when the embryogenic calli were transferred onto half strength WPM medium along with 2% (w/v) sucrose containing 2,4-D (0.1 mg/l), 2ip (0.05 mg/l), casein hydrolysate (100 mg/l) and L-glutamine (50 mg/l). The number of somatic embryo maturation (86.9) and germination (69.93) were produced on half strength MS medium (2% sucrose w/v) supplemented with abscisic acid (0.5 mg/l), BA (0.1 mg/l), GA3 (0.1 mg/l). Somatic embryo-derived plantlets were acclimatized and subsequently showed normal growth.
