Abstract

This study was designed to standardize the effect of picloram on plant regeneration from cotyledon explants of groundnut. High frequency of callus formation was noticed when the explants was cultured on MS medium with picloram. Three-week-old calli were transferred to shoot multiplication medium. Maximum number of shoots induced on half strength MS medium containing 3.11 uM picloram. The multiple shoots were separated and transferred to shoot elongation medium and BAP (8.88 uM) and NAA (2.69 uM) combination was found to be best for highest percentage of shoot elongation. The elongated shoot were transferred to rooting medium and the maximum percentage as well as number of roots was observed at 5.37 uM of NAA within 11 days of culture. Later the plants were transferred to small pots where the plants grew well.
