Abstract

Plant regeneration via direct organogenesis from apical shoot meristem of aseptically grown pigeonpea seedlings has been established. The protocol is simple, highly reproducible and shows regeneration frequency as high as 96.4%. This protocol has also been tested to be genotype independent. The efficiency of regeneration was found best for commonly grown genotype Bahar as compared to T-7 and ICPL 151. MS medium supplemented with different growth regulators along with 4% and 3% sucrose was found best for the induction of shoots (82.7%) and elongation of shoots (86.5%). The elongated shoots were efficiently rooted on MS medium supplemented with different concentrations of IBA. The best rooting response (100%) was obtained on MS medium supplemented with 0.3 mg/l IBA+3% sucrose. Rooted plantlets were successfully established in pots. The maximum survival (73.7%) of plantlets was obtained on a matrix having composition of sand: vermiculite (1:1).
