Abstract

A rapid and reproducible regeneration protocol has been established in Rosa indica via nodal shoot multiplication. Nodes of 0.5 cm long cultured on MS medium supplemented with IAA and BA produced multiple shoots. After 30 days the shoots began yellowing and were rectified by the use of AgNO3. A maximum of 53 shoots were produced per explant on MS medium containing 0.5 mg/l IAA, 1.0 mg/l BA along with 3 mg/l AgNO3 after three consecutive subcultures. Rooting of the regenerated plants were achieved in half-strength MS liquid medium augmented with 0.5 mg/l NAA and 2% sucrose. The plantlets of 4-5 cms were hardened in vermiculite and later transferred to field conditions where 90% survival rate was observed.
