Abstract

Mature embryos from 16 genotypes of Triticum aestivum and 4 genotypes of T. durum were cultured on MS medium with 4 modifications viz. MS2D (2.0 mg.1-1 2,4-D), MS 5D (5.0 mg.1-1 2,4-D), MSDC (5.0 mg.1-1 2,4-D + 100 ml.l-1 coconut milk) and MSDK (2.0 mg.1-1 2,4-D + 0.5 mg.1-1 kinetin). Considerable amount of variability was observed among genotypes, culture media and genotype x culture medium interactions. Embryos from the genotype JWJ 90-B-25 initiated maximum callus and exhibited maximum embryogenic callus formation. Medium MS2D responded maximum for these two traits; however, plant regeneration was lowest from callus cultures raised on this medium. Addition of coconut milk and 2,4-D to the basal MS medium (MS2D) regenerated maximum plantlets followed by MS5D, MSDK and MS2D.
