Abstract

An efficient plant regeneration system was developed for sorghum from different explants taken from germinating seedlings of three different varieties. The leaf base, the mesocotyl and macerated shoot apex were placed on Linsmaier and skoog(LS) medium with 2,4-Dicholorophenoxy acetic acid(2,4-D), with the cut end facing the medium. Callus was allowed to develop for 4 to 6 weeks and subsequently placed on medium containing different concentrations of Auxins and cytokinins. The medium with TDZ and Zeatin responded well in all the three explants. Multiple shoots were formed when shoot apex from five five-day-old seedling was placed on MS medium containing benzylaminopurine(BAP).
