An Efficient Protocol for Organogenic Callus Induction and Plant Regeneration in Tomato (Lycopersion esculentum Mill.)

Abstract
Organogenic callus induction and plant regeneration was achieved by using hypocotyls, leaf and cotyledonary segments. From the obtained different forms of callus, the green compact and nodular callus only produced the multiple shoots. More amount of pure organogenic callus only produced the multiple shoots. More amount of pure organogenic callus was obtained on the medium fortified with MS nutrients, 2.0mg/l IAA and 1.0mg/l KN. The induced organogenic callus produced multiple shoots on the same mother medium after 3-4 weeks culture. Induced shoots were also elongated on the same mother medium without the addition of any other plant growth regulators. Regarding multiple shoots, in cotyledon explants average of 9.6 shoots and in leaf explants 7.6 shoots were regenerated respectively. The elongated shoots were rooted on the medium fortified with 2.0mg/l IBA. The regenerated plants were successfully hardened on earthern pots after proper acclimatization.
