Abstract

Sesame (Sesamum indicum L.) is an important oilseed crop with high medicinal properties. Two commercial varieties (Amrit and Prachi) were used for direct and indirect organogenesis studies. Morphogenetic callus was induced in MS media supplemented with 2, 4-D (3.0 mg/l), NAA (2.0 mg/l) and BAP (1.0 mg/l). The callus when sub cultured in MS medium enriched with BAP (0.5 mg/l), KIN (1.5 mg/l) and NAA (0.5 mg/l) gave the highest response (68.46%) to shoot induction within a shorter period of 4.29 days. The juvenile shoots cultured in MS medium enriched with BAP (2.5 mg/l) and NAA (0.5 mg/l) produced the highest number and longer shoots. The internodal explants inoculated in MS media supplemented with BAP (3.0 mg/l), KIN (2.0 mg/l) and Adenine sulphate (2.0 mg/l) gave the best early response for direct organogenesis. For callus induction the seeds while for direct organogenesis internodes were found to be the best explants. Among the two varieties overall Prachi responds better to In vitro culture.
