In Vitro Culture of Inter-Specific Hybrid Ovules and Embryos of Cotton (Gossypium spp.)

Abstract
Post-zygotic in-compatibility is a barrier for inter-specific hybridization in cotton,therefore, in vitro inovuloembryo and embryo axes culture studies were carried out to recover crosses between cultivated dilploid and tetraploid and cultivated and wild species. Application of PGR'S particularly GA3 0.1percent in 2x X 4x crosses prevented abscission of crossed buds to the extent of 36.69 %. In Vitro inovuloembryoculture of 3 and 15 days old ovules revealed that embryo axes development was highest (13.38%) from 3 days than 15 days old (7.58 %), indicating the degeneration of embryo in in vivo. Embryo axes recovery was highest from ovules of 15-25 DAP, but the number of healthy plants from embryo axes obtained from 30-35 DAP was highest indicating in vivo condition is favourable for establishment of healthy seedlings. Presence of pentad & Hexad provided the cytological confirmation for intermediate nature of inter-specific hybrids.
