Abstract
To study the various effect of phytohormones on in vitro culture of Musa spp. cv. Yangambi an experiment was conducted at Banana Tissue Culture lab of Regional Plant Resource Centre, Bhubaneswar, Odisha, India. To develop a suitable protocol MS media was utilized supplemented with different concentrations and combinations of plant growth hormone (cytokinins and auxins) for this species. Observations were made through in-vitro culture based on different parameters like biomass (gm) and number of multiple shoot buds during different time interval and sub-cultured of explants from initial culture as well as multiplication culture stages. From various treatments explants cultured on induction medium containing MS + 4 mg/L BAP + 0.5 mg/L IAA + 80 mg/L ADS and sub-cultured on multiplication mediums supplemented with 3 mg/L BAP + 0.5 mg/L IAA + 0.25 mg/L NAA + 100 mg/L ADS, high biomass contents were obtained. The number of multiple shoot buds was significantly highest in medium containing MS + 4 mg/L BAP + 0.5 mg/L IAA + 0.25 mg/L NAA + 125 mg/L ADS and nearly similar observation was marked in MS + 1.5 mg/L BAP + 1.5 mg/L KN + 1 mg/L IAA + 0.25 mg/L NAA + 125 mg/L ADS.
