Abstract

With the advancement of molecular biology techniques many conventional genomic DNA extraction protocols has been developed for good, pure, high quality DNA with expensive and hazardous chemical reagents. Here we present a rapid, simple and efficient DNA extraction protocol using cetyl trimethylammonium bromide (CTAB) for orchid without involvement of liquid nitrogen and toxic phenols which are environmentally hazardous. The extracted genomic DNA was successfully used for Polymerase Chain Reaction (PCR) amplification of plant barcode genes (rbcL, matK and ITS) for orchid species.

Keywords: DNA extraction, CTAB, orchid, liquid nitrogen, phenol, PCR, barcode
